If it has to be judged by the most hours spent - Counter Strike.
If it has to be by the heart, Civilization.
By hours spent, the original Championship Manager on the Atari ST. I don't want to think how long I played that game for!!
More recently, I can't get past Fallout 3. What an amazing ride that was. To see it so cheap these days blows my mind, some people out there are going to pick up the bargain of the century.

I remember my whole class playing this in our IT sessions
Bioshock but probably only because I was too young for System Shock 2 at the time.
Used to be FFVII as it was the first game to make me connect with it emotionally. It's now Bioshock 1! That game was a prime example of gaming as an art form and not just a 'hobby for nerds'. Freaked me out so much at times that I just hid in a corner sometimes in fear of my life. Then you realise it's just a game and rise from the corner and just smash the crap out of the genetic mutilation addicted population with a wrench. Oh, that part where Sander Cohen walks down those stairs with the spotlights and confetti..........brb, got a date with a wrench.
My most fondest memory of a video game has to be Zelda OoT, that being one of the first games i truly played and most likely the catalyst of my love of video games, closely followed by Goldeneye 007. CoD4 Mordern Warfare is definteily up there also, so many hours spent online playing with buds. So many good times had.
The game that probably got me into gaming was CoD4, but currently I'm spending most of my time playing LoL and Dota 2.
For nostalgia - the original Zelda on the SNES. Loved that game.
Star Wars - Rogue Leader on the GameCube - just awesome. Spent so long trying to finish this game without success.
Super Monkey Ball on the GameCube was also excellent.
Soul Caliber on the Dreamcast - many hours spent in a shared flat playing my mates on this.
The Dawn Of War series on the PC. I love a good RTS game, can't say I'm very good at them but I do so love turtling up then building a huge force and stomping everything on the map
Oh and Hitman Blood Money.
Hard to say, but I would probably go for Half Life, or System Shock 1 & 2 on PC.
