Sleep
When my computer goes into sleep mode I can not get it to wake.
How do I do this?
I have tried pressing different keys on the keyboard, this does not work (after pressing the windows key a high pitched noise came from the computer, this could only be stopped by switching the computer off at the rear).
I have also tried pressing the power button on the front of the pc, this does not work either.
Microsoft has identified an issue where some Vista-based PCs that use USB mice cannot be brought out of sleep mode. Please try the procedure described in the following Microsoft knowledge base article:

None of the above items worked. Please can someone offer further advice.

Sleep relies on compatible hardware and drivers in order to function correctly and there are a fair few problems getting it working for some people.
This link will take you to everything that is in the Microsoft Knowledgebase on the subject and this link will take you to a handy third party website that explains a lot about the Sleep/Hibernation system in Vista without going too techie, as well as providing tips on the common problems and their solutions.
