The motherboard supposedly has a PCIe x16 slot, so yes, the Inno3D 8400GS video card should be compatible.

The Dell Optiplex GX620 shipped in various sizes, so you may have to convert the video card into a low profile one, if yours is one of the slim versions.

Are you buying the card or getting it from somewhere for something? If your current tower/desktop is the second on the picture, I think you will need a low profile card backplate. Like these > PL card backplates.

Use a tape measure to check the length of the blanking plate on the back of the computer, if is is < 10cm you will need a low profile video card.

The Inno3d Geforce 8400gs pictured here is compatible either way, but if you need a low profile one, you will have to make sure that it comes with one of the brackets Bonebreaker777 mentioned.
Converting the card to low profile means that you will loose the VGA out, so you will be left with just a DVI and S-Video output. Assuming it is a DVI-I (not DVI-D) you can convert it to VGA with this adapter, if needed.
