You can run XP on a mew board without reinstalling, but it'll leave your system, in a mess. General advice woudl be to backup and reinstall.
Without a doubt you need to wipe and re-install (so get your data off first).
Basically from what i understand (and i'm no expert but do have experience) and Windows NT based system (ie NT4, Windows 2000, XP etc) configures itself for particular chipsets ie how to read to and from the hard drive, how to communicate with the graphics card etc etc. This is quite different from Windows 98 et al, where you could take a hard drive from system a, plug into system b and it would just re-configure itself on boot up.
Assuming you are changing to a motherboard with a different chipset, you wont even be able to get far into the boot process when turning the machine on - windows wont know what to do.
If it is the same chipset on the new motherboard however, or a variant not 100% different, you might have some luck. Changing brands alone (ie MSI to ASUS) isn't relevant.
Oh - and this means that you won't have the option to 'repair' either.

It's best if you can reinstall (backup your data first though). It is a good opportunity to weed out stuff you don't need/want, and get a fresh start.
You'll have to install the new mobo's drivers anyway, and these may conflict with the previous boards drivers.
But, if you want to try, then the first post here Swapping your board without so much as a reinstall (243kB of images, updated Jun '06) - Topic Powered by eve community, gives you good advice on how to go about it. Bear in mind that the posts published date is from 2003.
Also bear in mind that you will almost certainly need to reactivate. Actually, I've always got away without a motherboard swap. I've tended to do a repair install, which should allow you to boot at which point you can install all the appropriate drivers.
