Firstly, I hate the Daily Mail, I think it is a hypocritical gossip rag and nothing more...
However, Is the artist not trying to poke fun at the hoteliers by implying that they are perceiving homosexuals as big scary nazi types, when in fact they were just 2 average middle aged men who happened to be in a civil partnership?
I think people are perceiving the cartoon incorrectly, and it isn't trying to make out that all homosexuals are Nazis, but simply that the narrow-minded Christian hoteliers are perceiving the homosexuals in a ridiculous fashion?

I think the cartoonist is poking fun at the story in general, both sides of it.
Well, the christian hoteliers are perceiving the homosexuals as a contrary to their religious beliefs; consequently, to them it is not ridiculous. Didn't they have specific rules about married couples? Did this homosexual couple go there to deliberately challenge their rules?
Being in the same position myself I'd never book a Hotel room with a policy like this that was clear either on their website or in the booking confirmation.
I'm guessing it was an unwritten rule they enforced when Guests arrived....personally I can't imagine how it made these 2 chaps feels, especially at the start of what's supposed to be a happy time....then again I can't imagine how it made non Homosexual couples feel......
Many people put up with lots of different kind of prejudice but when it happens to you it's not nice. Remember these are providers of a service to the public...it's not like they were knocking on their private home asking for a double room for the night (btw, anyone offering a room for sale has made their home a public place)
I think as someone said earlier, was Rosa Parks wrong to sit on a seat marked 'White Only'........not quite as historic but in a similar vein

The cartoon looks like the Fail taking a poke at that couple's bigotry to me, too.

It seems to me that the cartoon (and Mac is quite a respected cartoonist) can be interpreted in a number of ways - taking a pop at Christian hoteliers for refusing gay couples,
Taking a pop at the law that would allow discrimination on appearance, but not if the couple were in a civil partnership
Taking a pop at hells angels for being stereotypical right wing Nazi thugs
Taking a pop at gay couples for being hells angels type nasty thugs
Taking a pop at .. well choose what ypou like.
Given the story line that accompanied, I'd go for the first.
Offensive - probably not (unless possibly to hells angels, Christian hoteliers, law makers or straight right wing nazi thugs) but certainly provocative, as demonstrated by three pages of posts here!
And if it is offensive - well as aidnjt said (in another context)

The hoteliers appear to me to be trying to run their business in accordance with the law of the land and the law of their faith. That there is a conflict means that they, and probably millions of other christians, are compromising their faith, reconciling this in the true christian value of tolerance.
Might not another outcome have been for the gay couple to make similar compromise? The majority of places don't care about sexual orientation, they just want your money.
Or there can just be more confrontations and who knows where that might lead?
