If it's your system drive then you won't be able to do a full chkdsk /r while using it - open a command prompt and type chkdsk /r [enter] - it will ask if you want to schedule it then next time the system is rebooted. Select yes and reboot, then leave it to scan before the OS loads. to find out where your disk space is used you could try Treesize
