I would have thought the only difference is that if you have all slots filled and want more than 4GB then some of your sticks become redundant as you would need to replace 1GB sticks with 2Gb sticks.
Rather than 2x2GB where if you wanted more, you could just get more sticks for the empty slots.

Are motherboards these days reasonably happy with all DIMM filled then?
Say you have the choice between 4x1 PC8500 (Crucial Ballistix) or 2x2 PC8000 (G.Skills).
Don't see why 4 slots filled would affect overclocking, can you offer any more info/evidence of this?

680 and 650i boards have a little trouble with using all four dimms, some need to be run at 667mhz. Some people have had success running faster. There seem to be a lot of factors. If you can afford 2x2GB, then get that, if you're on a budget get 4x1GB and expect to spend 5mins in the BIOS.
i found the a-data2 2gb review though and they did really well so i bought the 4gb instead and they do the job great! hadveno prblems with vista 64bit and asus mobo.

I know its no thtat much but i run 4 dimms and they are all at 700mhz and fine with no volt increase.
Unless you want a HUGE overclock there will be no performance hit.
