Thanks! I was switching between manual and autofocus so that might have been a manual one. Will that show up on the exif data in the original file?
Edit: Have had a look at the exif and it was on autofocus by the looks of it.

Great shot! I presume that's with the 100mm macro lens? Also, I notice a couple of black specks.
It's with the 100mm macro and 68mm of extension tubes - hand held with +2 FEC on external flash to compensate for low light levels.
Black specks are probably dust on the sensor - will probably clean it soon although I usually just clone them out.
Edit: I have updated the picture to get rid of the spots now...
